Accurate prediction of soil N availability requires a sound understanding of the effects of environmental conditions and management practices on the microbial activities involved in N mineralization. We determined the effects of soil temperature and moisture content and substrate type and quality (resulting from long-term pasture management) on soluble organic C content, microbial biomass C and N contents, and the gross and net rates of soil N mineralization and nitrification. Soil samples were collected at 0–10 cm from two radiata pine (Pinus radiata D. Don) silvopastoral treatments (with an understorey pasture of lucerne, Medicago sativa L., or ryegrass, Lolium perenne L.) and bare ground (control) in an agroforestry field experiment and were incubated under three moisture contents (100, 75, 50% field capacity) and three temperatures (5, 25, 40 °C) in the laboratory. The amount of soluble organic C released at 40 °C was 2.6- and 2.7-fold higher than the amounts released at 25 °C and 5 °C, respectively, indicating an enhanced substrate decomposition rate at elevated temperature. Microbial biomass C:N ratios varied from 4.6 to 13.0 and generally increased with decreasing water content. Gross N mineralization rates were significantly higher at 40 °C (12.9 g) than at 25 °C (3.9 g) and 5 °C (1.5 g g–1 soil day–1); and net N mineralization rates were also higher at 40 °C than at 25 °C and 5 °C. The former was 7.5-, 34-, and 29-fold higher than the latter at the corresponding temperature treatments. Gross nitrification rates among the temperature treatments were in the order 25 °C >40 °C >5 °C, whilst net nitrification rates were little affected by temperature. Temperature and substrate type appeared to be the most critical factors affecting the gross rates of N mineralization and nitrification, soluble organic C, and microbial biomass C and N contents. Soils from the lucerne and ryegrass plots mostly had significantly higher gross and net mineralization and nitrification rates, soluble organic C, and microbial biomass C and N contents than those from the bare ground, because of the higher soil C and N status in the pasture soils. Strong positive correlations were obtained between gross and net rates of N mineralization, between soluble organic C content and the net and gross N mineralization rates, and between microbial biomass N and C contents.  相似文献

Previous studies have shown that soil fungal biomass increases towards more natural, mature systems. Shifts to a fungal-based soil food web have previously been observed with abandonment of agricultural fields and extensification of agriculture. In a previous field experiment we found increased fungal biomass with reduced N fertilisation. Here, we explore relationships between fungi, bacteria, N input and grassland age on real dairy farms in the Netherlands. We hypothesised that also in pastures that are still in production there is a negative relationship between fungal biomass and fertilisation, and that fungal biomass increases with grassland age in pastures that are still in production. We expected the fungal/bacterial biomass ratio to show the same responses, as this ratio has often been used as an indicator for management changes. We sampled 48 pastures from eight organic dairy farms. Sites differed in age and fertilisation rate. We determined fungal and bacterial biomass, as well as ergosterol (a fungal biomarker). Fungal and bacterial biomass and ergosterol, showed a negative relationship with N application rate, and correlated positively with organic matter percentage. In old pastures, fungal biomass and ergosterol were higher than in younger pastures. Because bacterial biomass responded in the same way as fungal biomass, the F/B ratio remained constant, and can therefore—in our data set—not be used as an indicator for changing management. We conclude that the changes in fungal and bacterial biomass were driven by changes in organic matter quality and quantity. The negative relationship we found between N application rate and fungal biomass adds to earlier work and confirms the presence of this relationship in pastures with relatively small differences in management intensities. Earlier studies on shifts in fungal biomass focused on ex-agricultural fields or restoration projects. Here we show that fungal biomass is also higher in older agricultural pastures.  相似文献

Soils are structurally heterogeneous across a wide range of spatio-temporal scales. Consequently, external environmental conditions do not have a uniform effect throughout the soil, resulting in a large diversity of micro-habitats. It has been suggested that soil function can be studied without explicit consideration of such fine detail, but recent research has indicated that the micro-scale distribution of organisms may be of importance for a mechanistic understanding of many soil functions. Current techniques still lack the adequate sensitivity and resolution for data collection at the micro-scale, and the question ‘How important are various soil processes acting at different scales for ecological function?’ is therefore challenging to answer. The nano-scale secondary ion mass spectrometer (NanoSIMS) represents the latest generation of ion microprobes, which link high-resolution microscopy with isotopic analysis. The main advantage of NanoSIMS over other secondary ion mass spectrometers is its ability to operate at high mass resolution, whilst maintaining both excellent signal transmission and spatial resolution (down to 50 nm). NanoSIMS has been used previously in studies focussing on presolar materials from meteorites, in material science, biology, geology and mineralogy. Recently, the potential of NanoSIMS as a new tool in the study of biophysical interfaces in soils has been demonstrated. This paper describes the principles of NanoSIMS and discusses the potential of this tool to contribute to the field of biogeochemistry and soil ecology. Practical considerations (sample size and preparation, simultaneous collection of isotopes, mass resolution, isobaric interference and quantification of the isotopes of interest) are discussed. Adequate sample preparation, avoiding bias due to artefacts, and identification of regions-of-interest will be critical concerns if NanoSIMS is used as a new tool in biogeochemistry and soil ecology. Finally, we review the areas of research most likely to benefit from the high spatial and high mass resolution attainable with this new approach.  相似文献

Maize straw and pea straw were added to five Pakistani soils from a gradient in salinity to test the following hypotheses: Increasing salinity at high pH decreases proportionally (1) the decomposition of added straw and (2) the resulting net increase in microbial biomass. In the non-amended control soils, salinity had depressive effects on microbial biomass C, biomass N, but not on biomass P and ergosterol. The ratios microbial biomass C-to-N and biomass C-to-P decreased consistently with increasing salinity. In contrast, the ergosterol-to-microbial biomass C ratio was constant in the four soils at pH>8.9, but nearly doubled in the most saline, but least alkaline, soil (pH 8.2). The addition of the maize and pea straw always increased the contents of microbial biomass C, biomass N, biomass P and ergosterol, but without clear effects of salinity. Highest mean contents of microbial biomass C and biomass N were measured at day 0, immediately after the straw was added. Straw amendments increased the CO2 evolution rates of all five soils without any effect of salinity. The same was true for total C and total N in the two fractions of particulate organic matter (POM) 63–400 μm and >400 μm. Lowest percentage of straw-derived CO2-C and highest recoveries of POM-C and POM-N were observed in the maize straw treatment and the reverse in the pea straw treatment. Yield coefficients were calculated for maize and pea straw based on the assumption that the balance gap between CO2 and the amount of POM can be fully assigned to microbial products.  相似文献

To improve soil fertility, efforts need to be made to increase soil organic matter content. Conventional farming practice generally leads to a reduction of soil organic matter. This study compared inorganic and organic fertilisers in a crop rotation system over two cultivation cycles: first crop broad bean (Vicia faba L.) and second crop mixed cropped melon-water melon (Cucumis melo-Citrullus vulgaris) under semi-arid conditions. Total organic carbon (TOC), Kjeldahl-N, available-P, microbial biomass C (Cmic), and N (Nmic), soil respiration and enzymatic activities (protease, urease, and alkaline phosphatase) were determined in soils between the fourth and sixth year of management comparison. The metabolic quotient (qCO2), the Cmic/Nmic ratio, and the Cmic/TOC ratio were also calculated. Organic management resulted in significant increases in TOC and Kjeldahl-N, available-P, soil respiration, microbial biomass, and enzymatic activities compared with those found under conventional management. Crop yield was greater from organic than conventional fertilizer. The qCO2 showed a progressive increase for both treatments during the study, although qCO2 was greater with conventional than organic fertilizer. In both treatments, an increase in the Cmic/Nmic ratio from first to second crop cycle was observed, indicating a change in the microbial populations. Biochemical properties were positively correlated (p < 0.01) with TOC and nutrient content. These results indicated that organic management positively affected soil organic matter content, thus improving soil quality and productivity.  相似文献

Summary The influence of surface growth of inoculated cyanobacteria (blue-green algae) on subsurface properties of a brown earth, silt loam soil was studied in reconstituted flooded soil columns. One blue-green algae species, Nostoc muscorum, become dominant within the first 7 days of inoculation. In light control columns (not inoculated) a bryophyte, Barbula recurvirostra, was dominant although significant growth of indigenous blue-green algae occurred. The blue-green algae counts were in the range of 1×106 g-1 dry soil in the surface layer (0–0.7 cm) in both columns. Any effect of surface phototrophic growth on soil properties was restricted to the surface layer. In inoculated columns there was a twofold increase in microbial biomass and an eightfold increase in bacterial numbers by week 13. However, bacterial numbers declined so that there was only a 2.8-fold increase by week 21. Dehydrogenase (x2.1), urease (x2.8) and phosphatase (x3.1) activities and polysaccharides (+69%) increased by week 21 as a result of the blue-green algae inoculation along with a significant improvement in soil aggregation. However, similar increases occurred in the light control columns, indicating that given appropriate conditions of light and moisture indigenous species may be ultimately as effective as introduced species in bringing about biochemical and microbiological changes to soil.  相似文献

Collembola and microbial biomass C were investigated in a field experiment with controlled agricultural traffic and crop rotation over a period of 27 months. The wheel-induced compactive efforts were applied according to management practices within the crop rotation of sugar beet, winter wheat, and winter barley. Increasing wheel traffic produced increasing soil compaction, mainly due to a reduction in surface soil porosity. Increasing soil compaction was accompanied by a decrease in microbial biomass C and the density of collembola. The influence of soil compaction on microbial biomass C was smaller than that of the standing crop. However, for collembola, especially euedaphic species, a reduction in pore space appeared to be of more importance than the effects of a standing crop. Within the crop rotation, microbial biomass C and the density of collembola increased in the order sugar beet, winter wheat, and winter barley.  相似文献

通过室内模拟试验,以不同C源、C/N比、水分及通透性为调控因子,对多氯联苯(PCBs)长期复合污染土壤的土著微生物强化修复进行了初步研究。结果表明,PCBs长期复合污染土壤中,在土壤水分含量为田间持水量的60%时,加入淀粉、葡萄糖和琥珀酸钠均在一定程度上增加了细菌和真菌数量,从而促进土壤中PCBs的土著微生物降解。不同种类的C源对PCBs污染土壤的土著微生物降解效果存在明显差异,且其降解效果与C源的施用剂量密切相关。当淀粉加入量为C1.0g/kg土时,土壤中PCBs的降解效果较好,而葡萄糖和琥珀酸钠加入量为C0.2g/kg土时,PCBs的降解效果明显。土壤C/N比为10:1的处理效果优于C/N比为25:1和40:1。土壤人为翻动有利于PCBs污染土壤中细菌和真菌的生长,提高土著微生物的代谢活性,从而促进土壤中PCBs的自然降解。这为进一步探讨加速土壤中PCBs降解的最适条件和研发POPs污染土壤的生物修复技术提供了科学依据。  相似文献

The long-term (9 years) effect of pig slurry applications vs mineral fertilization on denitrifying activity, N2O production and soil organic carbon (C) (extractable C, microbial biomass C and total organic C) was compared at three soil depths of adjacent plots. The denitrifying activities were measured on undisturbed soil cores and on sieved soil samples with acetylene method to estimate denitrification rates under field or potential conditions. Pig slurry applications had a moderate impact on the C pools. Total organic C was increased by +6.5% and microbial biomass C by ≥25%. The potential denitrifying activity on soil suspension was stimulated (×1.8, P<0.05) 12 days after the last slurry application. This stimulation was still apparent, but not significant, 10 months later and, according to both methods of denitrifying activity measurement (r 2=0.916, P<0.01 on sieved soil; r 2=0.845, P<0.001 on soil cores), was associated with an increase in microbial biomass C above a threshold of about 105 mg kg−1. The effect of pig slurry on denitrification and N2O reduction rates was detected on the surface layer (0–20 cm) only. However, no pig slurry effect could be detected on soil cores at field conditions or after NO3 enrichments at 20°C. Although the potential denitrifying activity in sieved soil samples was stimulated, the N2O production was lower (P<0.03) in the plot fertilized with pig slurry, indicating a lower N2O/(N2O + N2) ratio of the released gases. The pig-slurry-fertilized plot also showed a higher N2O reduction activity, which is coherent with the lower N2O production in anaerobiosis.  相似文献

Methane (CH4) oxidation potential of soils decreases with cultivation, but limited information is available regarding the restoration of that capacity with implementation of reduced tillage practices. A study was conducted to assess the impact of tillage intensity on CH4 oxidation and several C-cycling indices including total and active microbial biomass C (t-MBC, a-MBC), mineralizable C (Cmin) and N (Nmin), and aggregate-protected C. Intact cores and disturbed soil samples (0–5 and 5–15 cm) were collected from a corn (Zea mays L.)–soybean (Glycine max L. Merr.) rotation under moldboard-plow (MP), chisel-plow (CP) and no-till (NT) for 8 years. An adjacent pasture (<25 years) and secondary growth forest (>60 years) soils were also sampled as references. At all sites, soil was a Kokomo silty clay loam (mesic Typic Argiaquolls). Significant tillage effects on t-MBC and protected C were found in the 0–5 cm depth. Protected C, a measure of C retained within macro-aggregates and defined as the difference in Cmin (CO2 evolved in a 56 days incubation) between intact and sieved (<2 mm) soil samples, amounted to 516, 162 and 121 mg C kg−1 soil in the 0–5 cm layer of the forest, pasture and NT soils, respectively. Protected C was negligible in the CP and MP soils. Methane uptake rate (μg CH4-C kg−1 soil per day, under ambient CH4) was higher in forest (2.70) than in pasture (1.22) and cropland (0.61) soils. No significant tillage effect on CH4 oxidation rate was detected (MP: 0.82; CP: 0.41; NT: 0.61). These results underscore the slow recovery of the CH4 uptake capacity of soils and suggest that, to have an impact, tillage reduction may need to be implemented for several decades.  相似文献
